Serial intravascular staining reveals altered T cell trafficking in aged and SIV +rhesus macaques

چکیده
Abstract Lymphocyte trafficking is required for lymphocyte development and maturation, immune surveillance response, immunological memory. While integral a functional system, it may also play role in dysfunction. Patients with HIV the elderly have dysfunction characterized by increased inflammation, activation, susceptibility to infections cancer. As implicated these processes, we hypothesized that cohorts altered kinetics, resulting immunosenescence. To determine relationship between kinetics dysfunction, performed serial intravascular staining (SIVS) healthy, aged, SIV-infected rhesus macaques. SIVS uses sequential infusions of fluorescent antibody label cells vivo, enabling us follow them as they migrate over time. In all animals, found naïve central memory T underwent more faster into tissues while effector terminal were likely remain circulation. SIV +and aged observed subsets tissues. A possible result spend less time lymphoid tissue therefore be initiate an response. Increased cytotoxic entering potentially increasing hallmark immunosenesence both +patients. Using evaluate +rhesus provides new insights crucial system. Supported Intramural Research Program Vaccine Center, National Institute Allergy Infectious Disease, Institutes Health

Serial expertise of rhesus macaques.
Here we describe the development of serial expertise in 4 experimentally naive rhesus monkeys that learned, by trial and error, the correct order in which to respond to 3-, 4-, and 7-item lists of arbitrarily selected photographs. The probabilities of guessing the correct sequence on 3-, 4-, and 7-item lists were, respectively, 1/6, 1/24, and 1/5,040. Each monkey became progressively more effic...
متن کاملMechanisms underlying γδ T-cell subset perturbations in SIV-infected Asian rhesus macaques.
T cells that express the γδ T-cell receptor, which recognize microbial or stress-induced antigens, represent a minority of blood T cells but constitute a major proportion of intraepithelial lymphocytes in the gastrointestinal mucosa. As microbial products have been shown to translocate from the gastrointestinal tract into circulation in chronically HIV/Simian immunodeficiency virus (SIV)-infect...
متن کاملSocial behavior in aged rhesus macaques.
This study investigated the relationship between chronological age and social behavior in 42 adult male and female rhesus macaques (Macaca mulatta) in three specific areas: social interactions, social roles, and social networks. While both old males and old females share behavioral differences associated with senescence, this study reports that old females and old males are distinct in their so...
متن کاملSIV infection of rhesus macaques results in dysfunctional T- and B-cell responses to neo and recall Leishmania major vaccination.
HIV infection is characterized by immune system dysregulation, including depletion of CD4+ T cells, immune activation, and abnormal B- and T-cell responses. However, the immunologic mechanisms underlying lymphocytic dysfunctionality and whether it is restricted to immune responses against neo antigens, recall antigens, or both is unclear. Here, we immunized SIV-infected and uninfected rhesus ma...
متن کاملCollapse of Cytolytic Potential in SIV-Specific CD8+ T Cells Following Acute SIV Infection in Rhesus Macaques
Poor maintenance of cytotoxic factor expression among HIV-specific CD8+ T cells, in part caused by dysregulated expression of the transcription factor T-bet, is associated with HIV disease progression. However, the precise evolution and context in which CD8+ T cell cytotoxic functions become dysregulated in HIV infection remain unclear. Using the rhesus macaque (RM) SIV infection model, we eval...
